Question 5: Controlled vocabularies for Tissue tab [AF]

Description: Redundant and potentially confusing vocabulary in Tissue tab field related to preservation, i.e., Field Preservation (TisFieldPreservation_tab) and Initial Storage Medium (TisInitStorageMedium_tab). Could we change Field Preservation to be more of the PROCESS (mostly focused on temperature/cold chain process) of how the tissue was collected/stored IN THE FIELD would make more sense. Mapping tissue preservation process, type, medium, current storage to data aggregators such that outside folks can more effectively query our tissue holdings- see Question 5 above :) From what I can tell, other collections with large tissue holdings are mapping these data to DwC preparations field as “Liver (Frozen)” etc

Proposed Controlled Vocabulary (Field Preservation Method):

• Room temperature with buffer
• Room temperature without buffer (I can’t imagine folks doing this)
• Liquid Nitrogen
• Frozen on dry ice
• Refrigerated using ice packs
• Then keep current list related to MEDIUM used to preserve tissue for Initial Storage Medium Field, e.g., buffer, DMSO, ethanol or IMHO this field (Initial Storage Medium) should be restricted to ACTUAL MEDIA the tissue sample might have been placed into.
• Suggested Controlled Vocabulary (Initial Storage Medium):
• No buffer (dry)
• Unknown buffer
• Ethanol (with various concentrations)
• Ethanol (Unknown concentration)
• DNA/RNA Shield
• DMSO
• Glutaraldehyde
• Glutaraldehyde in Sorenson's phosphate buffer
• Lysis buffer
• Queen's Lysis Buffer
• RNAlater
