I want to detect moving object and their direction on my own video,but I did'nt have the calib file,how to use it on own video?

One possibility is to try single-frame calibration methods such as perspective fields or scalenet to get camera focal length and principal points (usually the center of images). Note this might not work well as single image calibration is ill-posed in many cases.
