2 new terms requested for parameters relating to transparent exopolymer particles, Jira ticket: BODCNVS-2556


Hello,

I would like to request some vocabs advice/new P01 codes for two Changing Arctic Ocean datasets please. These datasets contain two parameters:

Transparent Exopolymer Particle (TEP) Abundance L-1
Transparent Exopolymer Particle (TEP) Carbon µg L-1

Sampling protocol:

"Seawater was collected using a rosette sampler with Niskin bottles throughout the water column from 5 to 3,500 m where the depth allowed. At stations with a water depth shallower than 3,500 m, the water column was sampled to about 50 m above the seafloor."

Analytical protocol:

"TEP abundance and area were measured microscopically. The sample volume (10-250 ml) was chosen according to the prevailing TEP concentrations determined under the microscope onboard. Samples were filtered onto 0.4 µm nucleopore membranes (Whatman) and stained with 1 ml Alcian Blue solution (0.2 g l-1 w/v) for 3 seconds. Back at the lab, filters were mounted on microscopy slides that had been sprayed with white paint on the bottom side to avoid the filter pores becoming visible. Mounted filters were stored at -20°C. 30 images per sample were analysed using a DM 1,000 LED (Leica) equipped with a MC190 (Leica). The pictures with a resolution of 1388 x 1040 pixels were saved using Leica Application Suite Version 3.4.0 (Leica). All particles larger than 0.2 µm2 were analysed with ImageJ. A filter prepared with 10 ml MilliQ water served as a blank. The TEP-carbon content was calculated using the formula with D=-2.55 and r being the radius of the examined TEP: TEP-Cmicro [µg L-1] = 0.25x 10-6x r-D. "

Summary of discussion at weekly P01 catch-up:

  • Create both terms in GP_MODEL
  • Create new S29 - transparent exopolymer particle
  • For the carbon content of TEP parameter, suggestion was to use S06 Concentration as carbon, with new S29 transparent exopolymer particle.
  • new S03 filtration, staining
  • new S04 optical microscopy and image analysis
  • for the carbon content of TEP parmeter, new S05 - computation based on radius using formula from Mari (1999).

Proposed P01 pref lables:

Abundance of transparent exopolymer particle {TEP} per unit volume of the water body [particulate >0.4/0.45um phase] by filtration, staining, optical microscopy and image analysis

Concentration as carbon of transparent exopolymer particle {TEP} per unit volume of the water body [particulate >0.4/0.45um phase] by filtration, staining, optical microscopy and image analysis and computation based on radius using formula from Mari (1999)

@hanrev I am not sure we need to have TEP in the curly brackets within the P01 preferred label. The acronym can be included in the altlabel of S29 and in the altlabel of the P01 e.g. TEP_abundance and TEP_carbon_concentration or something similar.

  1. Abundance of transparent exopolymer particle per unit volume of the water body [particulate >0.4/0.45um phase] by filtration, Alcian blue staining, optical microscopy and image analysis
  2. Concentration as carbon of transparent exopolymer particle per unit volume of the water body [particulate >0.4/0.45um phase] by filtration, Alcian blue staining, optical microscopy and image analysis and computation using Mari (1999) formula

Proposed definitions:

For TEP abundance:
'Number of transparent exopolymer particles identified through Alcian blue staining and counted using optical microscopy and image analysis and normalised to a unit volume of water sample.'

For TEP-C concentration:
'The carbon content (mass) of transparent exopolymer particles, identified through Alcian blue staining and counted using optical microscopy and image analysis, computed from the volume equivalent spherical radius using the formula from Mari (1999) and normalised to a unit volume of water sample.'

For TEP abundance:
Pref. label = 'Abundance of transparent exopolymer particle per unit volume of the water body [particulate >0.4/0.45um phase] by filtration, Alcian blue staining, optical microscopy and image analysis'
Proposed definition 2: 'Number of transparent exopolymer particles retained by a 0.4 micron filter and identified through Alcian blue staining and counted using optical microscopy and image analysis and normalised to a unit volume of water sample.'

For TEP-C concentration:
Pref. label = 'Concentration as carbon of transparent exopolymer particle per unit volume of the water body [particulate >0.4/0.45um phase] by filtration, Alcian blue staining, optical microscopy and image analysis and computation using Mari (1999) formula'
Proposed definition 2 = 'Concentration expressed as the amount (mass or number of moles) of carbon in the transparent exopolymer particles retained by a 0.4 micron filter suspended in a unit volume of salt or fresh water and identified through Alcian blue staining and optical microscopy.'

For TEP abundance:

Proposed definition 3: 'Number of transparent exopolymer particles retained by a 0.4/0.45 micron filter, identified through Alcian blue staining, and counted using optical microscopy and image analysis, normalised to a unit volume of water sample.'

For TEP-C concentration:
