Computational chemistry

The Green site was identified by Dr Konstantin Popov (@kipUNC) and Prof Alex Tropsha at the UNC Eshelman School of Pharmacy.

Dr Popov also lead the virtual screening campaign that generated the initial hit and in addition supports the project with a computationally guided SAR-exploration - the second expansion was proposed from Enamine REAL was proposed by James Wellnitz at the UNC Eshelman School of Pharmacy, and Dr Popov.

Mutagenic studies

Confirming the Green site's conserved status through mutagenic studies was performed by Prof Ralph Baric at UNC Gillings School of Global Public Health, and Dr Vic Tse at St Louise University, School of Medicine.

Protein interaction - aggregation

The picogreen assay and GCI/WaveDelta assay for the intial selection of compounds for this project was done by Dr Tina Leisner in the lab of Dr Ken Pearce at the UNC Eshelman School of Pharmacy.

The picogreen assay was later moved to the labs of SGC in Toronto, where it is run by Dr Sumera Perveen, who also performs biophysical assays (SPR) and aggregation studies.

Antiviral activity - toxicity

The antiviral activity studies are performed by Dr Jessica Smith in the lab of Dr Alec Hirsch at the OHSU Vaccine and Gene Therapy Institute. Dr Smith also runs the Huh7-cell line toxicity tests.

Additional test on human cell lines were performed by Heather M Froggatt, Yeepeng Chan, and Audrey K Williams in the lab of Prof Ralph Baric at UNC Gillings School of Global Public Health.
